Technology Summary

This technology describes a novel class of Topoisomerase II (top2) inhibitors that are useful in treating cancer. Drugs that inhibit the top2 enzyme are among the most active anticancer agents discovered. However, many of the currently available inhibitors produce toxic side effects, have poor pharmacokinetics, or eventually become ineffective because malignant cells readily acquire resistance. Therefore, there is a need for developing new top2 inhibitor drugs that will overcome these limitations.

Azatoxin and its derivatives, which are derived by combining two parent compounds etoposide and ellipticine, are the first compounds rationally designed as inhibitors of top2. Azatoxins are also potent inhibitors of tubulin polymerization. These two anticancer activities can be successfully separated by synthesizing azatoxin derivatives to yield compounds which can be pharmacologically advantageous against tumor proliferation. The National Institutes of Health (NIH) is requesting public comment on draft guidelines entitled ``National Institutes of Health Guidelines for Human Stem Cell Research'' (Guidelines).

The purpose of these draft Guidelines is to implement Executive Order 13505, issued on March 9, 2009, as it pertains to extramural NIH funded research, to establish policy and procedures under which NIH will fund research in this area, and to help ensure that NIHfunded research in this area is ethically responsible, scientifically worthy, and conducted in accordance with applicable law. Internal NIH procedures, consistent with Executive Order 13505 and these Guidelines, will govern the conduct of intramural NIH research involving human stem cells.

The lack of appropriate labeling results in offlabel use of prescription drugs in many children and for many conditions. Contributing factors to this frequent offlabel use of drugs in pediatrics include the rarity of some conditions in children with limited patient availability, the ethical concerns regarding the conduct of clinical trials in children, the lack of accurate information about which drugs are used by children, and the lack of longterm data on the medications that are frequently used.

Background

The National Institutes of Health (NIH) Public Access Policy requires investigators funded by the NIH to submit, or have submitted for them, an electronic version of their final, peerreviewed manuscripts upon acceptance for publication to the National Library of Medicine's digital archive, PubMed Central, to be posted publicly within 12 months after the official date of publication. Congress required the NIH to implement this funding limitation in Division G, Title II, Section 218 of the Consolidated Appropriations Act of 2008 (``Section 218''). The Policy is intended to advance science, provide public access to the published results of NIHfunded research, and improve human health.

The current Public Access Policy is the culmination of years of effort and community interaction. Prior to passage of Section 218, the NIH undertook extraordinary public outreach concerning the issue of public access to the published results of NIHfunded research. Under the provisions of section 3507(a)(1)(D) of the Paperwork Reduction Act of 1995, the National Cancer Institute (NCI), the National Institutes of Health (NIH), has submitted to the Office of Management and Budget (OMB) a request to review and approve the information collection listed below. This proposed information collection was previously published in the Federal Register on November 3, 2008 (Volume 73, No. 213, p. 65387) and allowed 60 days for public comment. There was one public comment received which questioned why U.S. tax dollars are being spent on a study located in Europe. The investigator responded directly to the comment on 12/19/08 stating that this study costs less money to conduct in central Europe than in the U.S. since previous data has already been collected. Additionally, since this region has the highest rates of kidney cancer in the world a study in this area would provide a wealth of data in terms of the causes of kidney cancer.
